Cholinesterases in human skin.

چکیده
This study concerns the distribution of cholinesterases in human skin. Previous work on this subject was published last year by Hurley, Shelley and Koelle (1) These authors used a method originated by Koelle (2, 3). In Koelle's method, acetyithiocholine and butyryithiocholine are used as substrates. It is assumed that acetyithiocholine is split by specific acetyicholinesterase as well as by nonspecific cholinesterase; butyryithiocholine on the other hand is not split by specific acetyicholinesterase but is attacked only by non-specific cholinesterase. In Koelle's procedure any free thiocholine liberated in the section by enzymatic action from the thiocholine ester is precipitated locally as dupric thiocholine. On addition of ammonium sulfide the copper of cupric thiocholine transforms into brown insoluble cupric sulfide. This brown precipitate serves to mark the sites of cholinesterase activity. With this method Hurley, et al. (1) found specific cholinesterase activity around human eccrine glands. In my work, Gomori's (4) modification of Koelle's method was used.

Role of oligosaccharides in the pharmacokinetics of tissue-derived and genetically engineered cholinesterases.
To understand the role of glycosylation in the circulation of cholinesterases, we compared the mean residence time of five tissue-derived and two recombinant cholinesterases (injected intravenously in mice) with their oligosaccharide profiles. Monosaccharide composition analysis revealed differences in the total carbohydrate, galactose, and sialic acid contents. The molar ratio of sialic acid t...

متن کاملCatalytic Bioscavengers Against Toxic Esters, an Alternative Approach for Prophylaxis and Treatments of Poisonings
Bioscavengers are biopharmaceuticals that specifically react with toxicants. Thus, enzymes reacting with poisonous esters can be used as bioscavengers for neutralization of toxic molecules before they reach physiological targets. Parenteral administration of bioscavengers is, therefore, intended for prophylaxis or pre-treatments, emergency and post-exposure treatments of intoxications. These en...
متن کاملCholinesterases Enzymes Activities as Biomarkers of Farm Workers Exposed to Organophosphates in Two Communities of Khuzestan, Iran
Organophosphate (OPs) compounds are widely used in intensive agriculture to improve production, protect crops and control diseases vectors. The main mechanism of toxicity of OPs is the inhibition of the Cholinesterase enzymes. In this study the acetylcholinestrase (AChE), butyrylcholinesterase (BChE) and specific acetylcholinestrase (SAChE) activities as potential biomarkers of exposure to OPs ...
